Is there a way to use the ladder that the tunnel leads you to? I would think that it would lead to the main treasure room, but it was hinted that this could only be accessed from above.

No, it leads to the bottom of the elrvator to Ferro's armory. The reason you can't use it is when the elevator is down ( in Ferro's shop ) it blocks the shaft,
